Effects of Verapamil and Allopurinal on Ischemia-Reperfusion Injuy in Rat Liver

Abstract

|
|
Increased cellular calcium and the product of free radicals during ischemia and reperfusion appear to accelerate the cell injury in ischemic condition. It have been well known that verapamil, a slow calcium channel entery blocker, and
allopurinol,
xanthine oxidase inhibitor, may protect the various organ from ischemic injury. We induced warm ischemia of rat liver for 90 minutes and observed the biochemical(SGOT, SGPT, ALP, LDH and MDA) and histological changes after reperfusion. Also we
administered verapamil and allopurinol before ischemic insult to evaluate its protective effects. Serum level of hepatic enzymes and MDA were markedly elevated in the ischemia without medication and in the ischemia with allopurinol pretreatment.
The
level of hepatic enzymes and MDA in ischemia with verapamil pretreatment were significantly low(p<0.001). The level of MDA in ischemia with allopurinal pretreatment was slightly lower than the ischemia group without medication(p<0.01). On the
histologic
findings, severe centrilobular cell necrosis of the liver wa sobserved in the ischemia without medication and in ischemia with allopurinol pretreatment. In ischemia with verapamil pretreatment slight necrosis was observed we conclude that
protective
effect of calcium-blocking agent verapamil for ischemia-reperfusion-inducd liver injury was displayed, but not obviously with allopurinol.
